Die Entwicklung zweier Verfahren für die E. coli Fermentation eines rekombinanten Proteins wird in diesem Buch beschrieben. Durch den apparatetechnischen scale-up einer Schüttelkultur wirken einige Parameter nicht mehr limitierend auf den Fermentationsprozess. Darüber hinaus erweisen sich Medienzusammensetzung, Fütterungsrate der Nährstoffversorgung sowie Zeitpunkt der Expressionsinduktion des auf dem lac operon basierenden Expresionssystems als wichtige Parameter der Prozessführung. Erst wird eine batch-Fermentation in einem modernen komplexen Medium beschrieben. Bei der zweiten Methode handelt es sich um eine fed-batch-Fermentation in einem definierten Medium. Der Vorteil dieses Verfahrens liegt in der optimierten Ausnutzung des Metabolismus der Zelle. Der Metabolismus ist darauf ausgerichtet Energie für das Zellwachstum und die Zellerhaltung bereitzustellen. Bei der Expression des oftmals zellfremden Proteins bedarf es andere und mehr Metabolite. Durch die Abstimmung der Expressionsgeschwindigkeit auf die Wachstumsgeschwindigkeit der Zelle werden so Mangelerscheinungen unterbunden, sodass es nicht zur Zelllyse kommt und eine maximale spezifische Ausbeute erreicht werden kann.
